不同功能基因mRNA推断损伤时间的指标同质性

Homogeneity of Different Functional mRNA Indicators for Wound Age Estimation

摘要: 目的 探索大鼠骨骼肌挫伤后PUM2、TAB2、Cx45、CHRNA1四个不同功能基因mRNA表达在个体间的同质性高低。 方法 利用实时荧光定量反转录聚合酶链反应(quantitative real-time reverse transcription-polymerase chain reaction,RT-qPCR)检测上述PUM2、TAB2、Cx45、CHRNA1 mRNA的相对表达量,计算每个损伤组不同个体间的相对表达量的变异系数(coefficient of variation,CV),并比较CV极值、累积变异度、集中趋势(■CV)和离散趋势(CVCV)。 结果 PUM2、TAB2 mRNA在多个时间点出现较大CV,而Cx45、CHRNA1 mRNA的CV较小。累积变异度从大到小依次为PUM2 mRNA、CHRNA1 mRNA、TAB2 mRNA和Cx45 mRNA。PUM2 mRNA相对表达量的CVCV高于TAB2、Cx45、CHRNA1 mRNA的相对表达量(P<0.05)。TAB2、CHRNA1、Cx45 mRNA间相对表达量的CVCV差异无统计学意义(P>0.05)。 结论 参与生物过程调控作用的PUM2、CHRNA1 mRNA相对表达量个体间同质性最低,其次为TAB2 mRNA,参与细胞结构组成的Cx45、CHRNA1 mRNA相对表达量个体间同质性较高,在今后筛选用于损伤时间推断的mRNA指标时应注意其功能分类。

Abstract: Objective To explore the homogeneity level of four different functional mRNA (PUM2, TAB2, Cx45 and CHRNA1) expressions in rats with skeletal muscle contusion. Methods The relative expressions of PUM2, TAB2, Cx45 and CHRNA1 mRNAs were detected by quantitative real-time reverse transcription-polymerase chain reaction (RT-qPCR). The coefficient of variation (CV) of the relative expressions for different individuals in each injury group was calculated. The extreme value of CV, cumulative variability, and CVCV were compared. Results A high CV of PUM2 and TAB2 mRNAs appeared on several different time points. However, the CV of Cx45 and CHRNA1 mRNAs was relatively low. The cumulative variability from high to low was PUM2, CHRNA1, TAB2 and Cx45 mRNAs. The relative expression of PUM2 mRNA was significantly higher than that of TAB2, Cx45 and CHRNA1 mRNAs (P<0.05). There was no statistical significance (P>0.05) in the CVCV of the relative expression of TAB2, CHRNA1 and Cx45 mRNAs. Conclusion As the mRNAs involving in biological process regulation, PUM2 and CHRNA1 mRNAs show a lowest individual homogeneity of the relative expression followed by TAB2 mRNA. As the mRNAs participating in the composition of cellular structure, Cx45 and CHRNA1 mRNAs show a high individual homogeneity of the relative expressions. The functional classification should be considered for the screening of the mRNA indicators used for wound age estimation.
