Simultaneously Typing of Hp,Tf and Gc Phenotypes in Human Serum

Abstract: Hp,Tf,Gc are three genetic markers that are very important in individual identification and paternity testing in Forensic Science.In this paper,we report the method of simultaneously typing of Hp,Tf,Gc pheno- types in human serum by slab electrophoresis with polyarylamide gel. Before electrophoresis,the sample is mixtured with 5% Hb solution by 1:2.5(v/v).Electrophoresis is carried out at a contant voltage of 240V for three hours.After electrophoresis,the gel is stained with Benzidine for Hp phenotypes,then with Amino-Black for Tf and Gc phenotypes.The electrophoretogram of Hp,Tf,Gc shows in the cathodic,central and anodic region respectively.Using this method,we examine commonly the gene frequencies of Hp,Tf,Gc in human serum in Shanghai China.The obtained allele frequencies are as follow:Hp1=0.3182,Hp2=0.6818;Tfc=0.9886, Tfd=0.0076,Tfb=0.0038;Gc1=0.6667,Gc2=0.3333.The detection of three genetic markers in same gel plate is very effective and this method also available to the discrimnation of blood stain.We also discover that the slight hemolysis has little effect on the detection of Tf and Gc phenotypes.
