GlobalFiler?誖PCR扩增试剂盒验证及其STR遗传多态性

Validation of GlobalFiler?誖PCR Amplification Kit and the STR Polymorphism

摘要： 目的 测试GlobalFiler?誖PCR扩增试剂盒技术性能指标，评估其法医学应用价值，并对其遗传多态性进行调查。 方法 从灵敏度、混合样本、种属特异性、适应性、耐受性、一致性、均衡性、稳定性8个方面对该试剂盒进行测试，自动工作站提取北京地区汉族人群373名无关个体血样DNA进行扩增检测。 结果 GlobalFiler?誖PCR扩增试剂盒灵敏度较高，对混合、酶切降解和含抑制剂样品具有一定的检测分析能力，适应性和均衡性较好。21个STR基因座基因型频率分布均符合Hardy-Weinberg平衡（P＞0.05），PIC值在0.536～0.940，H值在0.558～0.933，DP值在0.783～0.992，PE值在0.243～0.874。 结论 GlobalFiler?誖PCR扩增试剂盒可用于法庭科学实际检案与建库，21个STR基因座在北京汉族人群中具有较高的遗传多态性，对法医学应用和群体遗传学研究具有重要意义。

Abstract: Objective To test the technical parameters of GlobalFiler?誖PCR Amplification Kit for its application to forensic application value and to investigate the genetic polymorphisms. Methods The validation was conducted in sensitivity, mixed samples, species specificity, adaptability, survivability, consistency, peak height balance and stability. The amplification and detection of the genomic DNA from 373 unrelated individuals from Beijing Han nationality were extracted by automation workstation. Results GlobalFiler?誖PCR Amplification Kit was adaptive to some mixed, degraded and inhibited samples. The power of sensitivity and adaptability and peak height balance showed well. The distributions of genotype frequencies for 21 STR loci in the population were all in accordance with Hardy-Weinberg equilibrium （P＞0.05）. The PIC value of the 21 STR loci was among 0.536 to 0.940; the H value was among 0.558 to 0.933; the DP value was among 0.783 to 0.992; the PE value was among 0.243 to 0.874. Conclusion GlobalFiler?誖PCR Amplification Kit is suitable for criminal cases and DNA database in forensic practice. And 21 STR loci in Beijing Han nationality have high polymorphism, which have application value in forensic practice and population genetics.
