    Expression of Caspase-8 after Brain Injury from Fluid Percussion in Rats

    Objective To provide the evidence of the relationship between brain injury and the time of injury. Methods Rats were contused on brain by fluid percussion, then were killed after injury for 15min, 30 min, 1,3,6,12 h, and 1,4,7,14 d respectively. The expression of caspase-8 were detected by immunohistochemical technology on rat brain section and the results were assessed by image analysis system in the cerebral cortex, thalamus, and hippocampus. Results The expression of caspase-8 in cortex and hippocampus could be detected in 30min after injury, increased significantly in 3h, reached apex in 1d after injury, remained 4d before decreased. In addition, the expression of caspase-8 can be detected in 1h after injury and reached apex in 1d after injury, and remained 4d then reduced. Conclusion It seems that the expression of caspase-8 should be a useful target for diagnosis of early brain injury.

    Objective To investigate the dynamics of the induction of S100β in different parts of rat brain following the diffuse brain injury. Methods Immunohistochemistry and auto-image analysis were to determine the expression of astroglial S100?茁 after diffuse brain injury in rats. Forty rats were distributed into groups according to injury time of 30 min, and 2,4,12,24 h, and 3,6 d after diffuse brain injury, and normal rats as control. Results The number of S100β positive cells in the four areas increased significantly followed by a decrease, and then a further increase. The expression of S100β could be detected increasing in 2h, and increased significantly in 4h, and it reached apex 12h after DBI, and decreased gradually to the level less than normal 3d, and returned to normal 7d following injury. In the postmortem injury groups, there were no significant changes in anti-S100β immunoreactivities in four areas of brain compared to the control group. Conclusion The present study showed the time-dependent expression of S100β is obvious following diffuse brain injury, and suggested S100β be suitable as a marker for brain injury age determination.

    Objective To study the relationship between DNA degradation and postmortem interval of corrupt corpse. Methods By determining the marrow DNA content with histochemical technique and image analysis. Results The content of marrow DNA decreased gradually with prolongation of postmortem interval, and it evencould be detected till 14 days after death. Conclusion There was a linear relationship between the degradation rate of the nuclear DNA and postmortem interval.

    Objective To study the relationship between postmortem interval and the myofibril fragmentation index of skeletal muscle. Methods Rabbit skeletal muscle were left in the room temperature for different postmortem intervals, and the protein concentration of each sample was detected by using biuret method. Furthermore, the myofibril fragmentation index of each sample was measured under the protein concentration level of 0.5mg/mL. Results The myofibril fragmentation index increased obviously according to the postmortem interval prolongation. Conclusion The myofibril fragmentation index may be used on the estimation of early postmortem interval.

    objective In order to improve the accuracy and reliability in sudden cardiac death, the pathogenesis and relationship between the viral myocarditis and dilated cardiomyopathy were investigated. Methods Improved immunohistochemical technique was adopted to detect the expression of the dystrophin in myocardium from 25 viral myocarditis, 28 dilated cardiomyopathy and 17 control cases including normal, coronary atherosclerotic heart disease and hypertension heart disease as control. Results The positive rate of dystrophin protein expression in control group was 100%, that in viral myocarditis was 88%, and that in dilated cardiomyopathy was 57%, There were significant differences among three groups (P<0.05), and the correlation between viral myocarditis and dilated cardiomyopathy group(r= -0.526)were also found. Conclusion The myocardial cytoskeletal protein is disrupted in viral myocarditis and dilated cardiomyopathy ,and the dystrophin protein may be involved in the pathogenesis of viral myocarditis and dilated cardiomyopathy. The viral infect and impair heart functions by cleaving host dystrophin proteins may ultimately contributes to the viral myocarditis to the converting from dilated cardiomyopathy.

    Objective To find out the pathological change and the toxic mechanism of Chloranthus serratus Roem. et Schalt in mice. Methods Mice were intoxicated by oral administration with extracts of Chloranthus serratus Roem. et Schalt followed by pathological, serum biochemical, and coagulation mechanism examination. Results The LD50 in mice was 41.12 g/kg; All poisoned mice serum BUN and ALT increased markedly; Thrombocyte decreased and coagulation time increased; The organ index of liver, spleen and kidneys increased significantly; The cells of liver, kidney and heart were degeneration and necrosis, There were extensive hyperemia and hemorrhage in many organs. Conclusion The experiment suggests that the target organs were liver, kidney, heart and blood vessels; The toxic mechanism was the damage on the mitochondrional, endoplasmic reticulum and coagulation system.

    Objective To seek the pathomorphological targets for forensic expertise in anaphylactic shock. Methods The expression of IgE in hearts, lungs, livers, spleens, kidneys, gastrics, intestinals, tracheas and tonsils of anaphylactic shock guinea-pigs was observed at 0, 6, 12 h and 24 h respectively by tissue chip S-P immuno-histochemical method. Results Positive expression of IgE presented in lungs and tracheas in the test group with the peak at 0 hour and it declined as time advanced, and also there were significant differences at different times(P<0.05). Conclusion The immuno-histochemical method of detecting the expression of IgE in lungs, tracheas and spleens can be supposed to be the pathomorphological targets for forensic expertise in anaphylactic shock. The weakening of the positive expression of IgE in lungs and tracheas as the time advanced suggested that in this kind of case the autopsy should be arried out as early as possible.

    Objective To observe the changes of cardiac functional indicators in spontaneously hypertension rats (SHR) suffered by struck injuries of soft tissues, form which to discuss the significance in relationship between injury and disease. Methods SHRs were divided into four groups according to the degree, and whose thigh was struck with the homemade hit instrument. Then the blood pressure, heart rate as well as serum biochemical indicators (CK, CK-Mb, LDH, Cr, BUN, HBDH and AST) were examined. In addition, the morphological changes of heart, brain and kidney were also observed. Results The observe values of heart rate and blood pressure of SHRs seemed have relation to the degree of damage, but there were no statistically significance between different hit degree groups. In terms of serum biochemical indicators, all seven indicators of the hit group had significant difference with that of the control group, and the AST?CK?CK-MB levels were difference between the group of different strike degree. Conclusions Struck injuries of soft tissues can result in forepart damage of cardiac muscle. The biochemical indicators reflect the degree of injury, and they could be used as a type of objective enzymology indicators for analysis of the relationship between injury and disease in forensic practice.

    Objective To investigate expression of ryanodine receptors (RyRs) in rabbit penile corpus cavernosum smooth muscle(CCSM) cells. Methods New Zealand White Rabbit CCSM cells were cultured by primary tissue culture. Using CCSM cells and fibroblast have different adherence velocity, CCSM cell can be purified. Identification of CCSM cell was by inverted microscope and immunofluorescence technique. The cDNA sequence of RyRs was found out by searching genebank. Three pair of primers were designed by Primer Premier 5.0. The RyRs subunits mRNA was detected by reverse transcription PCR in cultured CCSM cells. Results After 7 d, we found growth of cultured cells. While 15~20 d, cells filled the bottom of culture flask. They were identified by inverted microscope and immunofluorescence technique. After purification, purity of CCSM cells was near to 100%. It suggested only RyRs1 subunit was expressive in CCSM by RT-PCR. Conclusion RyRs1 subunit is expressed in CCSM cells. It suggests that RyRs contribute to the regulation of Ca2+ in CCSM cells.

    Objective To study identical indexes of frontal sinus in different locative computer radiology. Methods Frontal sinus radiographs were analysed by combining indexes of kinds of distances and patterns, further using descriptive study to describe frontal sinus and to analyse data statistically. Results The 8-identical classification of frontal sinus was proposed, basing on the ratio of frontal sinus width and orbit width, the bilateral asymmetry, the superiority of side, the numbers of arches of upper borders(left and right), the partial septa(left and right)and the location of the central septa. There was no statistical significance of sinus between sexes(P>0.05). Conclusion Frontal sinus is tremendous inter-individual variation so it can be used for identification, but it can not be used for sex determination.

    Objective To discuss the coding of full dentition with 32 locations and measure the characteristics of some bony indexes in occlusal-facial digital radiology(DR). Methods To select randomly three hundred DR orthopantomogram and code the full dentition, then analyze the diversity of dental patterns. To select randomly one hundred DR lateral cephalogram and measure six indexes (N-S,N-Me,Cd-Gn,Cd-Go,NP-SN,MP-SN) separately by one odontologist and one trained forensic graduate student, then calculate the coefficient variation(CV) of every index and take a correlation analysis for the consistency between two measurements. Results (1)The total diversity of 300 dental patterns was 75%.It was a very high value. (2)All six quantitative variables had comparatively high CV value.(3) After the linear correlation analysis between two measurements, all six coefficient correlations were close to 1. This indicated that the measurements were stable and consistent. Conclusion The method of coding full dentition in DR orthopantomogram and measuring six bony indexes in DR lateral cephalogram can be used to forensic identification.

    Objective To solve the difficulties of identification of Sarcosaphagous flies such as Lucilia sericata (Meigen) and Lucilia cuprina (Wiedemann) which could not be identified by analyzing the 278 bp and 635 bp regions of the gene encoding for cytochrome oxidase subunitⅠand Ⅱ(COⅠand COⅡ) in mtDNA. Methods Specimens were collected from the corpses of rabbits on the grassland in Huhhot and Chengdu, the sequences of 551 bp region of 16S rDNA of their mtDNA were analyzed, the multiple-alignment program DNAMAN(version 4.0) and MEGA 2.1 sofeware were employed for sequence alignments neighbour-joining tree construction. Results Lucilia sericata (Meigen) and Lucilia cuprina (Wiedemann) were distinguished successfully by sequence analysis of The 551 bp region of the gene of 16S rDNA. Conclusion The 551 bp region of the gene of 16S rDNA of sarcosaphagous flies can be used for identifying them on species level effectively. It is likely to be a successful compliment to identify the sarcosaphagous flies by sequence analysis of COⅠand COⅡin mtDNA.

Conclusion Several factors were involved in the produce of STR slippage, such as amount of modulate DNA, concentration of MgCl2, property of DNA polymerase, motif sequence of STR loci, sample, etc.

    Objective To establish a method for whole genome amplification (WGA) based on (multiple displacement amplification MDA), and achieve DNA analysis of the human genome from low copy number (LCN). Methods DNA sample was used for WGA according to the REPLI-g kit protocol (QIAGEN,Germany). WGA product was used for DNA analysis according to the Applied Biosystems Profiler Plus kit protocol (ABI, USA). Results WGA product of DNA sample (10 pg) can be used for STR genotyping. Conclusion WGA technology could be helpful for LCN DNA analysis.

    Objective To analyze mitochondrial DNA (mtDNA) polymorphisms in coding area and provide a theoretical basis for applying in forensic science. Methods The primers of 8162F/8483R and 13070F/13299R were designed according to the Anderson′s sequence. Using PCR-sequencing method to detect polymorphisms of mtDNA nt8162-8483 and nt13070-13299. Results The lengths of the amplicons were 322 bp and 230 bp respectively. There were 24 mitochondrial haplotypes defined by 21 variable positions in both regions. The gene diversity was estimated at 0.751 1, and the probability of two randomly selected individuals having identical mtDNA types was 0.256 4. Conclusion The polymorphic sites within mtDNA coding area can be useful in combination with mtDNA control region in order to increase the discrimination power.

    Objective The possibility for the identification of GHB administration through hair analysis was investigated to provide method and information for toxicology examination of GHB. Methods A GC/MS assay for GHB in hair was developed. Endogenous levels of GHB in hair, time course of GHB in hair, relationship between GHB levels in hair and hair color or administration dose were also established by guinea pig model. Results Endogenous levels of GHB in guinea pig black hair and human black hair were (3.01±1.41) ng/mg (n=28) and (1.02±0.27) ng/mg (n=20), respectively. GHB levels in black hair were increased by GHB administration and related with drug dosage, and also much higher than in brown and white hair. Conclusion Analysis of GHB in hair is suitable for investigation of GHB abuse in forensic toxicology and GHB level in segmental analysis compared with endogenous level of GHB may provide useful information about GHB administration.

    Objective Liquid chromatography-tandem mass spectrometry(LC-MS/MS) was developed for the determination of opiates in biological samples according to the emerging problem in drugs abuse. Methods Opiates such as heroin, 6-acetylmorphine, morphine, codeine, acetylcodeine, hydrocodone and hydromorphone were isolated from human blood, urine, oral fluid and hair using a simple extraction and consequently analyzed using LC-MS/MS. The method was evaluated by real cases. Results The mobile phase give the optimum separation for opiates. The detection limit of morphine in urine with dilution and liquid-liquid extraction and in hair is 10ng/mL, 0.01 ng/mL and 0.01 ng/mg, respectively. Conclusion The method is simple and rapid, offering superior sensitivity and selectivity for opiates. The target compounds comprising hydrocodone and hydromorphone enlarge the applied area.

    Objective To investigate the distribution of GHB in biological fluids and tissues and provide method and information for detcetion of GHB in Biological Fluids and Tissues Method The concentrations of GHB in Biological Fluids and Tissues were analyzed by gas chromatography-mass spectrometry; Two groups of rats were fed with 1000 mg/kg of GHB and were killed at 1 hour and 3 hours post dose, respectively. Results GHB concentrations of the administrated groups distributed in a degressive order of urine, stomach, blood, intestine, kidney, lung, spleen, heart, liver and brain. Conclusion Urine was the best specimen for the determination of GHB concentrations in biological fluids and tissues.

    Objective To explore the criminological characteristics of mental retardation (MR) in forensic psychiatry. Methods The record scale of forensic psychiatric assessment designed by ourselves was used to analyse the criminological characteristics of 83 offenders with MR, and to compare the criminological characteristics of mild MR with that of moderate and severe MR. Results The mild MR accounted for 62.7%, moderate and severe MR was 22.9%. The percentage of sex offenders in MR was 37.3%, manslaughter 34.7%, property offences 28.0%, respectively. Additionally, 96.1% cases with MR have definite criminal motives, and the criminal history was established in 34.7% cases. Significant differences of criminal premeditation (χ2=11.9,P=0.001), criminal aim(χ2=7.531, P=0.006), criminal motive(χ2=7.920, P=0.019) and criminal types(χ2=7.855, P=0.02) were found between the mild MR and the moderate, severe MR. Conclusions The criminal offenders were mostly found in mild MR. The sex offenders and manslaughter were in outright majority, and most of them had definite criminal motives. The proportion of offenders in mild MR who had criminal premeditation and criminal aim was higher significantly than which in the moderate, severe MR. The proportion of offenders in moderate, severe MR whose criminal motive was for sex was higher than that in mild MR.

    Objective To discuss the relation between degree of body impairment and that of thoracolumbar spinal injuries resulting from road traffic accidents, and sum up the experiences in body impairment assessment and its regularity. Methods For comprehensive body impairment assessment, 477 cases of thoracolumbar spinal injuries in road accidents have been sorted out, reassessed and rediagnosed. In addition,analyses have been undertaken about their treatment, the assessment of the degree of their thoracolumbar dysfunction,nerve dysfunction and the relations between injuries and sequelaes. Results The analyses show that the degree of thoracolumbar dysfunction and that of the post-injury nerve dysfunction don′t necessarily depend on the quantity and degree of spinal injuries. However, the position suffering from the thoracolumbar spinal injuries has an immense impact on the thoracolumbar dysfunction, and the nerve impairment result mainly from the T1-L1 spinal injuries. The research also shows that there has been a high misdiagnosis rate in hospital about the spinal injuries. Conclusion In body impairment assessment, the cause and effect relations between the injury and degree of injury extent should be analyzed, the injury extent should be employed as principal evidence, and the degree of spinal dysfunction should be taken into greater consideration.

The author collected and analyzed 49 cases of these tangles from many aspects, such as sex and age of the dead, hospital,information of autopsy, fault of medical action and so on,and discovered the normal causes of death,medical action's effects and the causes of tangle. It would be useful to the forensic appraisal,settlement and prevention of these medical tangles.

    Lumbar sacralization and lumbarization are congenital spine malformation.16 cases with lumbar sacralization or lumbarization were reviewed. Through studying the etiopathogenisis and clinical manifestation we analyze the relationship between injury and disease. 6 cases of 16 have lumbar sacralization. 10 cases are attributed to lumbarization. Most of this cases have backleg pain more or less. As a forensic doctor we suggest that pay more attention to this problem in our identify practice and locate the sequence of vertebral body more exactly.

    Objective To study the extracting and genotyping method of sweat latent fingerprint samples involved in cases. Method Chlex 100 extraction method was used to extract DNA. STR loci were typed after PCR amplification by Profiler Plus kit. Results All the sweat latent fingerprint samples involved in cases obtained reliable results of STR genotyping. Conclusion It is very important to find and extract sweat latent fingerprint samples properly for STR genotyping.

    Objective To analyze cause of medical accidents and actuality of technical identification in medical tangle. Methods 32 cases(17 death,15 survive) of medical tangle by technical identification (according to sex?age?mostly diseases ?sequel ) and identified results( whether or not mistake?cause and effect connection?duty degree)have been studied. Results 13 cases of 32 medical accidents have been determined.19 cases have been attributed to no medical accidents. Causes of medical accidents were most due to negligence of sense of duty. Conclusion The incidence rate of medical accidents can be decreased by strengthen colligated stuff of medical affairs personnal. We suggest that our state bring out more perfect legislation of autopsy in order to gain positive effect of technical identification in medical tangle. The medical mistake among grade of medical accidents should be added so that justice of identification could be improved.

The traditional methods for detecting menstrual blood were reviewed and their obstacles in forensic application were discussed. Matrix metalloproteinase-11 is a kind of protease which degrades the extracellular matrix. Some researches had indicated that matrix metalloproteinase-11 might be a new marker for specifically and sensitively detecting menstrual blood. The structure, function of matrix metalloproteinase-11 and its application in menstrual blood identification were reviewed.
