    Preliminary Application on Test Paper of Gold-labelled Antibody against Fibronectin EⅢA Splicing Variant

    Objective To develop the test Paper of gold-labelled antibody against fibronectin EⅢA splicing variant, which can be available in forensic wound interval estimation. Methods Two sensitive antibodies were compared with enzyme link immunoabsorband assay (ELISA). After colloid gold labeled, the effects of the two antibodies were tested by methods of Dot immunogold filtration assay and gold immunochromatography assay, respectively. The test paper was finally appraised by applied in experimental skin injury in rats. Results On the test paper, detected line appeared in three hours wound age group, and then the darkness of positive staining became darker with injury time prolonging, while control normal skin cannot found to be positive staining. Conclusion The gold-labeled test paper can be useful in estimation of wounding interval in forensic science.

    Objective To study the value of Fibronectin(Fn) immunohistochemical staining for diagnosing slight viral myocarditis. Methods The heart samples of human with myocarditis were studied by using LSAB immunohistochemical staining with anti-fibronectin antibody. Results Dense deposition was found in the myocardium of human with myocarditis. Some Fn-positive cardiomyocytes were observed. Conclusion Slight degeneration of cardiomyocytes could be identified by Fn-LSAB immunohistochemical staining and Fn-deposition is one of the reliable marks for inflammation in the myocardium.

    Objective To explore the relationship between the pink teeth phenomenon and the cause of death as well as its significance in forensic medicine. Methods Inspection method was adopted to observe the pink teeth phenomenon in different causes of death. Ten rats were selected for every experimental groups, which were then divided into two groups:Eight in fresh group with teeth pulled immediately, and two in decayed group with body decayed in water firstly. The teeth pulled from rats were immersed in 75% alcohol and observed at different immersion time. Results In every fresh groups, pink teeth phenomenon was not observed when they were pulled immediately, whereas it emerged gradually after the teeth immersed in 75% alcohol, and the color showed distinct and constant four hours later. In decayed groups, Pink teeth phenomenon was observed immediately when teeth pulled, it became distinct and constant after one hour′s immersion in alcohol. So it was more distinctive in the decayed groups than that in the fresh groups. Conclusion There is no significant connection between the pink teeth phenomenon and the cause of death, thus it may not be subject to forensic identification.

    The Expression of GFAP after Brain Concussion in Rats

    objective To study the expression of GFAP and pathologic changes after rats brain concussion, so that to provide evidence on brain concussion for forensic identification. Methods Forty-five SD rats were divided into 3,6,12,24 h and 2,4,7,10 d and normal control groups in terms of different wounding time after brain concussion model established, and the expression of GFAP after rats brain concussion were then observed by using SP immunohistochemical method. Results In normal control brain, low-level GFAP expressions could be observed. After six hours′ brain concussion, GFAP positive cells increased obviously. The trend reached to the peak at 7d, partly declined at 10 d, then decreased gradually. Conclusion Brain concussion induced the expression of GFAP. The detection of GFAP could be useful for diagnosis of brain concussion on forensic pathology, and could be a reference index for timing of injury after brain concussion

    Objective To study cellular mechanism of cardiomyocytes injury in the early stage of crush injury by observing some effects of crush injury rat sera on cultured neonatal rat cardiomyocytes. Methods One to three days old neonatal rat cardiomyocytes were cultured in vitro and some effects of crush injury rat sera on beating rate, cell surface area, total protein content, 3H-Leu incorporation, intracellular calcium concentration ([Ca2+]i) and Fos protein expression were observed in cultured rat cardiomyocytes. Results Compared with normal rat serum group, crush injury rat sera decreased beating rate(beats/min) of cardiomyocytes from 88.3 to 26.4, cell surface area, total protein content, 3H-Leu incorporation, [Ca2+]i(nmol/L) and PI of Fos protein expression were increased. Conclusion Crush injury rat sera suppress cell beating, increase intracellular calcium, induce Fos protein synthesis and cause cell hypertrophy, which may cause cardiac injury in the early stage of rush injury.

    Objective To identify sarcosaphagous flies and their larvae, pupa. Methods sarcosaphagous flies and their larvae, pupas were collected from human corpses and their surroundings in the Weifang city. A 304 bp region in COI gene was analyzed by mtDNA sequencing. Results The studied region showed no sequence divergence within same species and significant difference were found between different species in all samples. Conclusion It is a practical approach to identify these Sarcosaphagous flies and their larvae, pupas by sequence analysis of the 304 bp region of the COI in mtDNA.

    Objective To study the relationship between the expression of c-Fos protooncogene and skeleton muscle contusion of rats, and to search for a sensitive marker of timing for skeleton muscle contusion. Methods Fitty Sprague-Dawley rats were randomly distributed into control group and experimental groups. The expression of c-fos was microscopically observed by immunohistochemical method. Results With the time prolonged the c-fos positive intensity and area were increased. Positive expression of c-fos protein appeared at 15min after skeleton muscle contusion, and reached to the peak at 1h after skeleton muscle contusion, then decreased gradually and returned to the normal level on 1d after skeleton muscle contusion. Conclusion The detection of the expression of c-fos protein could be a sensitive marker for timing skeleton muscle contusion.

    Objective To explore the differences in Haversian system between human and animal bones through imaging analysis and morphology description. Methods Thirty-five slices grinding from human being as well as dog, pig, cow and sheep bones were observed to compare their structure, then were analysed with the researchful microscope. Results Plexiform bone or oeston band was not found in human bones; There were significant differences in the shape, size, location, density of Haversian system, between human and animal bones. The amount of Haversian lamella and diameter of central canal in human were the biggest; Significant differences in the central canal diameter and total area percentage between human and animal bones were shown by imaging analysis. Conclusion (1) Plexiform bone and osteon band could be the exclusive index in human bone; (2) There were significant differences in the structure of Haversian system between human and animal bones; (3) The percentage of central canals total area was valuable in species identification through imaging analysis.

    Objective To investigate the changes of excitability of rats′ gastrocnemius after rats were died from mechanic asphyxia, so as to make an objective evidence for estimating of the early postmortem interval. Methods After rats were sacrificed by mechanic asphyxia, the reaction of rats′ gastrocnemius to variable electric stimulation at different period after death were recorded by electricphysiological method.Results Changes in the properties of excitable tissue to electric stimulate in rats′ gastrocnemius after mechanic asphyxia seems to be well correlated with the early postmortem interval and appeared a linear relation in certain period. Conclusion The regular changes of excitability in muscle might be a useful means for estimating of early postmortem interval.

    Objective To study different viewpoints to the conclusion and treatment,And the following up factors concerning social effect after the judgment of forensic psychiatry. Methods By letters,calls,visits and by investigation forms made by myself, 208 testimonied cases were followed up separately from judicial organ or other organizations and institutes ,individuals testimonied or their family members,victims and their family members. Results Most testimonied individuals were married man of 30 years old or so whose arerage education were 4.84 years and testimonied by the public security organs. In criminal cases, violental criminals (129 cases, 83.77%)were much more than non-violental criminals(25cases, 16.23%) and homicide criminals(44cases,28.57%) were most common in the former but civil cases were few. The rate of retstinvony was 2.93%.The testimonied individuals and their family members thought the condemn were appropriate (76.47%) for the people without criminal capacity and thought the condemn were appropriate (41.94%) or not appropriate(41.94%)for the people with criminal capacity. The opinions of the condemn for partial criminal capacity were between the former two cases. And they thought the condemn for 28 cases of non-guilty were appropriate (71.43%) and not appropriate(10.7%).7 victims were dead. 10 victims were crippled. 10 victims restored to health. In 41 persons testimonied and set free with a verdict of "not guilty",4 homicided again (4.87%),2 set on fire (4.87%),2 stealed (4.87%) and 3 had wrecked behavior (7.31%). Conclusion So it is suggested that the department concerned should keep criminals with mental disorder under control and treatment.

    Objective Studying forensic medical problem related with RTA leading PTSD and supplying accumulating evidence for psychiatric compensation in court. Methods One hundred and fifty six victims of RTA were recruited who applied to court for a costs order. The victims were examined for psychiatric diagnosis by psychiatrists and for rank of impairment by experts in forensic clinical medicine. The self-report psychopathological status and quality of life were also measured. Results Eighty one victims of 156 (51.92%) fulfilled the criteria for PTSD (ICD-10). Morbidity difference in male and female were significant; The more serious extent of impairment is, the more PTSD′possibility is; The scores in World Health Organization Quality of Life were lower and in SAS and SDS were higher in PTSD group than in non-PTSD group. Acquirement of awarded costs could obviously prevent PTSD. Conclusion The higher PTSD incidence existed in the RTA victims who applied to court for a costs order, and acquirement of awarded costs could obviously prevent PTSD.

    Objective To develop a PCR-based STR system for genotyping of 18 loci (Amelogenin, D3S1358, vWA, FGA, D8S1179, D21S11, D18S51, D5S818, D13S317, D16S539, TH01, TPOX, CSF1PO, D7S820, D2S1338, D19S433, D12S391 and D19S253). Methods By using primers labeled with four color fluorescent (FAM, HEX, TAMRA and ROX), two multiplex amplification reaction systems were developed to genotype Amelogenin and 17 STR loci. Results Amelogenin and these 17 STR loci were genotyped successfully in different kinds of biological samples by the kit. Conclusion The STR amplification kit developed in our study gives a new approach to genotype these 18 loci in a efficient, steady and reliable way.

    The Application of PCR-SSCP in Forensic mtDNA Typing

    Objective To study the application of PCR-SSCP in forensic mtDNA typing. Methods Primers flanking the mtDNA HV-I and HV-II regions were designed. By PCR-SSCP techniques, 70 family trios and 140 unrelated Wuhan Han individuals were investigated and analyzed. Results In 70 family trios,the SSCP profiles in region HV-I and HV-II of children were not same to that of their fathers in 98.57% and 97.13% respectively but were identical with their mothers. In 140 unrelated Wuhan Han individuals, 21 haplotypes were found in HVI, GD=0.9556; 16 haplotypes were found in HVII, GD=0.9356. Conclusion PCR-SSCP technique may be useful in forensic mtDNA typing, especially for screening the suspects.

    Objective To investigate the genetic polymorphism of 12 STR loci in 240 Han population and evaluate application of the 12 STR loci systems in forensic cases. Methods 12 STR loci were amplified with Muti-loci STR PCR system, which were genotyped with electrophoresis separation on dPAGE. Results 12 STR loci system had high power of discrimination(DP) and it was suited for analysis of dated bloodstain especially. Conclusion The 12 STR loci systems is simple economy and practicality, which is useful in forensic identification & disputed paternity.

    Objective To establish a fluorescent multiplex PCR system for typing Y-STR loci Y-GATA-A7.1, DYS456 and DYS443, and investigate their haplotype frequencies in Chinese Han population. Methods 203 unrelated males of Han population living in Zhengzhou were typed by fluorescent multiplex amplification system and ABI 3100 genetic analyzer. Results In Zhengzhou Han population, 5,6 and 6 different alleles were observed for Y-GATA-A7.1, DYS456 and DYS443 loci, and their gene diversity (GD) were 0.669 2, 0.583 9 and 0.705 3 respectively. A total of 44 different haplotypes formed by these three loci was identified and the haplotype diversity (HD) reached 0.952 3. Conclusion The fluorescent multiplex system for these three Y-STR loci will be very powerful for forensic individual identification and paternity testing in Chinese Han population.

    Objective The aim of the present study was to establish a rapid and robust assay used to simultaneously genotype SNPs by the single nucleotide primer extension (minisequencing) with the SNaPshot Kit and obtain the population genetic data in Chinese population in Sichuan. The analysis of single nucleotide polymorphisms (SNPs) is a promising application in forensic casework. Methods 12 Y-SNPs, which were SRY2627, SRY1532, M13, M20, SRY8299, Tat, M69, M9, 92R7, M17, M19 and M112, were multiple amplificated and the PCR products were pooled , Purified, and then used as templates for the minisequencing reaction with the commol/Lercially available SNaPshot Kit. Then the products of minisequencing reaction were detected by capillary elcetrophoresis. Results 78 genomic DNA individual samples from Sichuan, China and 5 semen stain samples from sexal criminal scene were analyzed and two haplotypes could be identified. Conclusion A rapid method has been established to analyze the 12 Y-SNPs by multiplex PCR and minisequencing. It can be applied in forensic casework successfully.

    Objective To develope an easy to use, rapid and accurate test for detecting buprenorphine based on the principle of competitive immunoassay. Methods Monoclonal antibody against buprenorphine was conjugated with colloidal gold and dispensed on the glass fiber. The complete antigen Buprenorphine-BSA and the goat anti-mouse IgG polyclonal antibody were separately sprayed on the nitrocellulose membrane as the test line (T line) and the control line(C line). The rapid test kit was the final assembled product of test strip with the plastic cover. Results A total of 100 urine samples were tested for buprenorphine by immunochromatographic and GC/MS methods. the accuracy was 99.0%. It is found the test kit can only detect by cross reaction with other 45 kind drugs. Conclusion Rapid test kit can detect buprenorphine in the samples in 5 minutes. The cut-off value of the test is 100 ng/mL.

    Three cases of newborn emphysema were reported. Parenchymatous emphysema, interstitial emphysema and bullous emphysema were all found in the three cases. One of the three cases showed accumulation of air in interstitial tissue. The air penetrated into septa of the lung at the beginning, then it probably split the hilus pulmonis to reach mediastinum as well as subcutaneous tissues of the chest, neck, abdomen, inguinal region and scrotum. The oxygen inhalation under pressure during first-aid may be the cause of the newborn emphysema.

Methods the autors investigated 43 head trauma children and 50 normal children, using Wechser Inrelligence Test(C-WISC). Data were analyzed by SPSS10.0 on computer. Results head trauma children had lower FIQ,VIQ,PIQ than normal children. Pervaded head trauma has more contribution to the damage of the children′s intelligence.

    Objective To investigate forensic diagnosis application of three-dimentional reconstruction with spiral computed tomography in fracture of anatomical complicated bones. Methods Selected eleven patients of bone fracture who were examined with SCT 3D and conventional X-ray examination. The location, number and characteristics were observed and analyzed. Results In all of eleven patients with bone fractures, X-ray examination could detect thirty-four rib fracture, one scapula fracture,two nasal fracture, one metacarpal bone incomplete fracture and one left tibia-fibula fracture, one pubis fracture. While there were forty-seven rib fracture, one scapula smash fracture, one nasal fracture with obvious displacement and eliminate one misplace, one left tibia-fibula obsolete fracture and one sacroiliac joint dislocation, one No 5 lumbar vertebrae pedicle of vertebrae arch fracture. Combining 3D reconstruction images, coronary and sagittal reconstruction images could show clearly the fracture line, location of fracture, number of fracture, displacement and recovery. Conclusion 3D reconstruction technique of SCT is a very useful examination method in the objective forensic diagnosis of anatomical complicated bones fracture, it excels the routine X-ray examination.

    Objective To found the quantifiable index of "The severe degree of negligence" in describing the general severity degree of medical malpractice or medical dispute. Methods "The severe degree of negligence" can be calculated by the way of multiplying the coefficient of medical malpractice′s grade by the coefficient of responsibility degree. Results There are 15 grades of "The severe degree of negligence" through calculation, from the severest degree of 1 to the lightest degree of 20. Conclusion "The severe degree of negligence" can give an order of severe degree to different grade and different responsibility of medical malpractice. According to this order, the operation of medical malpractice and medical dispute settle will be easier and more rationality.

    In recent years, mental disability estimation are more than ever in the practice of forensic psychiatric expertise. The standards about mental disability estimation in the Disability Evaluation Criteria of Traffic Accident Injury are ambiguous and difficult to operate, it cause to different understanding in one case. This article discuss some common questions through 3 aspects in mental disability expertise according to some typical cases, there are the first, we should compare the IQ score before and after injurious in intelligence estimate. The second, the mental disability estimation should be done at least one year after the end of medicine treatment. The third, mental estimate scale should be used in mental disability estimation.

    As the most popular and important inherited marker used in forensic identification, short tandem repeat (STR) always have partial DNA profiling or even no results when handling degraded or minute DNA sample. Through redesigning primers close to STR core repeats, MiniSTR can access shorter STR loci and increase success rate of DNA profiling in degraded or minute DNA sample. The review provides an update on the advancement of miniSTR research to give information in the practice of forensic science.
