    PCR-based analysis of the D1S80 locus and its application in paternity test

    Allelic data for the DIS80 locus was obtained by usingthe PCR and subsequent analysis with high - resolution horizontal PAGE technique. In a sample of 184 unrelated Chinese, the D1S80 locus had 75 phenotypes and 20 allelic.The distribution of genotype was in agreement with expectedvalues according to the Hardy - Weinberg equilibrium. Analysis of the D1S80 locus by the PCR has been successfully applied in 150 cases of questioned paternity.

    DNA was extracted from human bones remained for 3 to15 years by using the method developed in our laboratory. Apair of X - Y homelogous primer were used to amplify thespecific DNA fragments of Amelogenin gene. All the 35 sensitivples yielded correct results. Our method is rapid, sensitiveand reliable for the sex determination of skeletal remains.

    F13A01 FESFPS and vWA were extracted and amplifiedusing denaturing polyacrylamide gel electrophoresis and silverstain. The distribution of allete frequencies of F13A01, FESFPS and vWA loci in Jingpo ethic group living in southwestern Yunnan province. were investigated. It was observed thatthere were 8 alleles and 13 genotypes in F13A01 locus; 7 alleles and 18 genotypes in FESFPPS locus; 7 alleies and 21genotypeS in vWA lous.

    Determination of chloroquine in human urine

    A GC method was established for qualitative and quantitative determination of chloroquine in human urine. The chloroquine is extracted from 2ml urine with 2 x 2ml cyclohexame: chloroform (8: 2). The retention time of chloquine is9.44min. The linear of quantitation is 0 - 50μg /ml. Therecovery of the procedure in 25μg g/ml is 87 .0%, CV = 7.9% (n = 5). The limit of the determination is 200ng/ml.The method can also be used in blood. A case is reportedhere. In that, we detected chloroquine in urine and blood.The quantitation of chloroquine in urine and blood is 0. 475mg/ml and 3 .68μg/ml, respectively. A metabolite of chloroquine - desethylchloroquine is also found in the urine.

    A GC/ECD method was established to determine triazolam in human urine qualitatively and quantitatively. The triazolam was extracted from 2ml urine with 2 x 2ml chloroform.The retention time of triazolam was 10. 74 min. The limit ofthe determination was 0. 5ng/ml. The recovery of the procedure in 5 ng/ml was 95.98%, CV = 7. 85% (n = 5). Thelimit of quantitation is 2 - 50ng/ml.

    8 allele and group specificity at HLA - DRB1 locuswere genotyped in 159 Unrelated individuals of Liaoning Han,with polymerase chain reaction - sequence specific primers.The range of gene frequencies was from 0 .02201 - 0 .23899.Heterozagosity was 83%. 33 out of 36 theoretical genotypeswere identified and the result conformed to Hardy- Weinbergequilibrium law. The present method can be used not only inpaternity test, individual identification, but also in clinicgraft matching between donor and recipient as well as thestudy of HLA.

    Ultra - stucture changes of autolysis of hepatocyte werestudied using transmission electron microscope(TEM). It wasfound that autolysis Started 0. 5 hour after death, with theheterochromatin of nuclei shrinking abnormally and smooth endoplasmic reticulum expanding. The changes developed todistinction after 1h, and progressed continuously after 2h,3h, 4h and 5h. It was found that (1) the mitochondria wereswelling, its matrix density decreasing and flocculent densebodies (FDB) emerging; (2) the rough endoplasmic reticulum and perinuclear cistern were expanding. It was suggestedthat ultra - structure changes of the autolysis be correlated tothe postmortem time.

    46 cases of medical disputes were analyzed. The causesof medical disputes were analyzed and the features of forensicidentification and medical compensation were discussed. Results showed that 37 cases had causality (cause and effect relation) between medical treatment and negative results. It is concluded that forensic medical identification can play an important role in the civil compensation of medical disputes.

    23 cases of thrombus formation in cardiac cavity werestudied. 9 were caused by extensive trauma in soft tissue, 13by various cardiac diseases. There was no significant difference between trauma and heart disease (p > 0. 05). Thewhite thrombus formation was mainly caused by truma andwas often found in the right cardiac cavity (P< 0.001 ). Themixed thrombus was mainly caused by heart disease and nodifference was found between in fight and left cardiac cavity(p > 0. 05). The types, positions and causes of thrombuswere significantly related (p < 0. 001 ). The causes, pathologic mechanism and forensic identification of thrombus formation in cardiac cavity were discussed.

    Pathological changes due to intoxication of tetramethylenedisulphotetramine (TETS ) were studied. The resultsshowed that the signs of asphyxia were obvious, so were thecongestion and edema of the brain. The spotty and focalhemorrhages in brain stems, multiple myolysis of papillarymuscles and contractionbands necrosis of myocardium wereobserved occasionally. The main clinical symptom of intoxication was freqUunt clinic - tonic convulsions, which weresimilar to the grandma epilepsy. The poisoned died mainly in3 hours. The general poisoning symptoms, LD50, toxicological mechanism and medicolegal expertise were also reviewed.
