The use of microwave for immunohistochemical technology in forensic pathology ZHOU WEI;ZHU JIAZHEN;WANG FEI (SUPREMEPEOPLE`S PROCURATORATE;PRC;BEIJING 000) 1997, 0(4): 193-194+.  Abstract ( 1548 )   A microwave oven (MWO) of National brand wasapplied for Labeled Streptavidin Biotin immunohistochemical staining (LSAB method) on autopsymaterials of 20 cases to detect 10 kinds of antigen ofheart and brain :The keys to manipulate MWO asfollows: (l) Recovery of antigen: submerge sectionsin 0.01M citrate buffer (pH 6.0), turn MWO' to theposition of DEF (210W) for 10 min, reachingtemperature 92 C -98 C .(2) add normal serum, firstantibody, second biotinylated antibody and streptavidin complex, then turn MWO to the position ofLOW (70W) for 5 min, reaching temperature 20 C37 C. Other Steps were the same as the standards.Results showed' MWO can recover the destroyed orcovered antigen, improve positive staining degree,reduce background's stain and shorten staining time. Macro，micro and ultrastructural observations of pituitary, adrenal, pancreas and spleen in experimental brain death WEI BENQING;LI DEXIANG;WANG DEWEN (THE PEOPLE'S MIDDLE COURT OF LINYI CITY;SHANDONG;00) 1997, 0(4): 195-198+.  Abstract ( 1941 )   The purpose of this study is to observe the macro,Micro and ultrastructural changes of the pituitary, adrenal, pancreas and spleen in the experimental primary brain death. The results showed: 1. Acinar zymogen granule of pancreas decreased, karyopyknosis of islet cells formed 2.dilation and blood stasis of spleen sinus happened 3.adrenocortical karyopyknosis formed, mitochondrial crista vanish.4.Pituitary karyopyknosis and cytolysis developed. 5.There was correlation between the extent of lesions And time after brain death. By the SYSTEM the photos of ChineseG banding chromosomes were analyzed. It showedthat the karyotype of the chromosomes of personswas recorded in high speed and efficiency, and thatthe chromosomes of alike ordinal number fromdifferent persons could be compared directly. Thatwould presage, by this kind of the high effectiveoperation, a favorable condition was created for theapplication of the polymorphism of the chromosomesto the parentage diagnosis. GUANGFUXIRD;SHANGHAI 000J) 1997, 0(4): 200-202+.  Abstract ( 1917 )   The methods using microwave irradiation for rapidpreparation of acetyl, trifluoroacetyl, pentafluoropyl,heptafluorobutyl and MSTFA derivatives ofMorphine, AMP and MAMP were developed.Conventional techniques for the reaction mixture tobe heated need 30 min at 60-70"C to formderivatizations, but by microwave irradiation, only2-3 min is needed. The mass spectral fragmentationpatterns and gas chromatographic retention times ofthe derivatives obtained by both microwaveirradiation and conventional heating are similar. In this paper, a model 1090HPLC with DAD and a zorbax ODS column wasused. The mobile phase was methanol: water: tritelyamine (75:24.7:0.3) with PH 7.5. Cyproheptadinewas used as internal standard in this method. Bloodsamples were extracted with the solid-phaseextraction method and the liquid-liquid method.
