吗啡戒断大鼠中脑腹侧背盖区GAP-43的表达变化

Expression of GAP-43 in Midbrain Ventral Tegmental Area of Morphine Withdrawal Rats

摘要: 目的 观察吗啡戒断大鼠中脑腹侧背盖区生长相关蛋白-43(growth associated protein-43,GAP-43)在不同时程中的表达,探讨GAP-43在吗啡戒断记忆中的作用。 方法 采用腹腔递增注射盐酸吗啡并自然戒断的方法建立吗啡依赖1周、2周和4周戒断模型,运用免疫组织化学染色观察中脑腹侧背盖区GAP-43的蛋白表达,并用Image-Pro Plus 5.1图像分析软件对结果进行分析。 结果 中脑腹侧背盖区GAP-43的蛋白表达随依赖时间的延长表现为先降低再升高(P<0.01)。 结论 GAP-43可能在中脑腹侧背盖区参与了吗啡戒断记忆。

Abstract: Objective To observe the protein expression of growth associated protein-43 (GAP-43) in midbrain ventral tegmental area in morphine withdrawal rats at different time, and to evaluate the effect of GAP-43 on morphine withdrawal memory. Methods Rat models of morphine dependent 1 week, 2 weeks and 4 weeks were established by morphine hydrochloride intraperitoneal injection with increasing doses to establish natural withdrawal. The protein expression of GAP-43 in midbrain ventral tegmental area was observed by immunohistochemical staining and the results were analyzed by Image-Pro Plus 5.1 image analysis system. Results With prolongation of dependent time, the expression of GAP-43 was decreased then increased in midbrain ventral tegmental area. Conclusion GAP-43 could play a role in morphine withdrawal memory in midbrain ventral tegmental area.
