摘要: 目的用测序方法验证12个STR基因座的基因型。方法根据各STR基因座的特异性序列,对CSF1PO、FGA、TH01、TPOX、VWA、D5S818、D7S820、D8S1179、D13S317、D16S539、D18S51和D21S1112个STR基因座设计了PCR引物并对标准品9947A和突变样本进行PCR产物测序。结果标准品9947A和突变样本的测序结果均与其基因分型结果一致。结论建立的测序方法准确、灵敏,可以用于STR基因型的确认。

Abstract: Objective To validate the genotype of 12 STR loci by sequencing method. Methods According to the special sequence of 12 STR loci(CSF1PO, FGA, TH01, TPOX, VWA, D5S818, D7S820, D8S1179, D13S317, D16S539, D18S51 and D21S11), the relative PCR primers were designed. PCR products of control DNA(9947A) and STR mutation samples were sequenced. Results The sequencing results of both the control DNA(9947A) and STR mutation samples were consistent with their genotyping results. Conclusion The sequencing method developed are accurate and sensitive, can be used for the validation of STR genotyping results.
