Determination of sex using PCR of the X-Y homologous gene amelogeuin

Abstract: Sex determination is carried out by simultaneouslyamplifying a X-specific and Y-specific DNA frag-ments which are 106hp and 112hp longrespectively in one PCR reaction using one pair OfPrimers which are SPecific to the first intron Of theX-Y homoIogous gene AmeIogenin.PCR productsare Separated on a PAGE electroPhoresis followedby visulization using silver stain.All the bloodStain.muscle,saliva and hair samples gave correctsex determination.Less than or as little as 50pgtemplate DNA and the 16-year-old blood stain allcan achieve satisfactory amplification.The resultsshowed that the method discribed in this paper is arapid,sensitive,reliable and easily manipulated sextest highly adapted to forensic SPecimen especiallythose petrified and degraded.
