    Analysis of Band Types of Human and Animal Hemoglobin before and after Treatment with PCMB by Isoelectric Focusing

    Hemolysates of 41 kinds of animals including human,fetal and monkey's one,were studied by isoelectric focusing on PAG Plate (pH3.5-9.5).Species featuces of Hb band pattern,ability for identification and alternation of Hb band pattern after treatment with P-Chloromercuribenzonate (PCMB) as well as its influence on the species identification of Hb were disscused as well.Approaching to the application of IEF for identifying species gave the following results:Class of animals can be identified hy characteristic distri- bution of the band pattern;Human Hb may be distinguished from that of other animals but monkey's one of which band types somewhat difficultly discriminated because of similar band type,Differences of band patterns are also shown between animals,which may be used for identification.Aftry treatment with PCMB,both human and monkey's Hb are broken down to and β subunit.The different PI value of α subunitin both human and monkey's Hb can be used for identification and much easily to distinguish human Hb from that of the other animals.

    This paper shows the detection of the phenotypes of isoenzymes EsD and GLOI for fresh blood cells using the same mixed starch agarose gel electrophoresis and Tris-maleic acid buffer system at pH7.4.Among 302 fresh blood samples,the EsD gene frequencies were:EsD~1=0.641,EsD~2= 0.359;Among 401 fresh blood samples,the GLOI gene frequencies:were: GLOI~1=0.1895,GLOI~2=0.8105,It is useful to paternity,test in Nangjing area.

    12 monoclonal antibody (McAb) include 5 anti-A,5 anti-B,1 anti- H and 1 anti-A+B,and polyclonal human serum anti-A andanti-B were test in parallel with more than 20 animal erythrocytes include pan troglodyles pango pygmaeus,monkey,cynoeephalus balouin etc.We found some McAb can discriminate all the measuing animals,that make it possi- ble to use McAb in species identification.McAb anti-A,anti-B and anti-H using in forensic sicence have two adventages:a)high specifi- city,some of them can discriminate primate animals.b) it can finish species identification and ABe groups measure in one procedure.

    It was presented that the results of EsD typing of Chinese population in Guangzhou area using agarose gel eleetrophoresis.Among 600 unrelat- ed healthy individuals tested,the numbers of EsD 1-1,EsD 2-1 and EsD 2-2 types were 163(27%),303(50.5%)and 135(22.5%),respectively, The gene frequencies of EsD~1 and EsD~2 were 0.5225 and 0.4775.It was consistent with Hardy-Weinberg equilibrium.The period of storage of bloodstains which could be typed correctly lasted 2 weeks from May through July in Guongzhou.The blood samples which were left in room condition and decaied naturally could be typed for 9 days.

    A method was described for determining erythrocyte acid phosphatase (EAP) phenotypes from fresh human blood and bloodstain by thin-layer polyacrylamide gel isoelectric focusing (PAGIF),obtaining arrow,even, clear isoenzyme bands Some of the advantages of thin-layer PAGIF were increased resulation and dried bloodstain for twomonthes can be detected. Using this method,We detected the EAP phenotypes from 283 unrelated individuals in Shanghai.The results were as follows:EAP A 4.24%,EAP BA 28.98%,EAP B 66.78%;p~a=0.187,p~b=0.813;Did not find p~c.The discriminating power (DP)of EAP was 0.47.

    This study was shown that the Esterase isoenzyme in rat liver would change after its death,especially within 6 hours.The changes of Ester- ases in rat liver appeared regularly.It was found that some ons decreas- ed.and some ones increased.We thought this method was very useful in determining the time of death.

    The HLA-AB typing of human spermatozoa identified by two colour fluorescence technique is reported in three cases.with the acridine orange and the ethidium bromide as stains,the human spermatozoa affected by HLA-AB antibody appeared two distinct colour fluorescence: the orange-red indicated dead spermatozoa earring the relevant HLA-AB antigen and jade green living ones noncarrying the antigen. Applied to the identification in disputed paternity,the HLA-AB typ- ing of human spermatozoa as haploid would raise the nonfatherexclusion probability (NFEP) of the assaying system,and as well as raise the paternity index (PI) and the relative chance of paternity (RCP).

    The identification and recognization of textile fibers by using differ- ential scanning calorimetry (DSC) technique is deteiled.In this study we mainly applied the DSC to identify the fibers dyed with a type of dye in different concertration and identify a kind of dye in different type.The results are showed that:a,the DSG method can be used to identify the dyes in the same or similar colour but in different types,bs the DSC curves of dyed fibers appeared not only the fibers special peaks but also the new special peak that dyes act on fibers.
