血液中常见除草剂的UPLC-HRMS分析

Analysis of Common Herbicides in Blood by UPLC-HRMS

摘要: 目的 建立一种血液中10种常见除草剂(百草枯、敌草快、草甘膦、草铵膦、氰草津、莠去津、吡草胺、乙草胺、氯磺隆、甲磺隆)的筛查及定量方法。 方法 以乙腈-水溶液[V(乙腈)∶V(水)=3∶1]为蛋白沉淀剂,用超高效液相色谱-高分辨质谱法(ultra-high performance liquid chromatography-high resolution mass spectrometry,UPLC-HRMS)对血液中的10种除草剂进行检测。 结果 10种除草剂在各自线性范围内线性关系良好(决定系数R2≥0.993),回收率为67.4%~111.9%,相对标准偏差为1.5%~10.8%,准确度为85.1%~106.1%,日内精密度为2.7%~13.5%,日间精密度为3.3%~13.3%。 结论 该方法操作简便、回收率高,可同时对血液中多种除草剂进行快速准确的定性筛选和定量分析。

Abstract: Objective To develop a method to screen and quantify 10 common herbicides (paraquat, diquat, glyphosate, glufosinate, cyanazine, atrazine, metazachlor, acetochlor, chlorsulfuron, and metsulfuron) in blood. Methods With acetonitrile-water solution [V(acetonitrile)∶V(water)=3∶1] as protein precipitant, 10 common herbicides in blood were detected using ultra-high performance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS). Results All the 10 herbicides had good linearity in their linear range (coefficient of determination R2≥0.993), with the recovery rates 67.4%-111.9%, the relative standard deviations 1.5%-10.8%, the accuracies 85.1%-106.1%, intra-day precisions 2.7%-13.5%, and inter-day precisions 3.3%-13.3%. Conclusion This method is easy to operate with high recovery rates. It enables rapid and accurate qualitative screening and quantitative analysis of various herbicides in blood simultaneously.
